急性腹泻病患者与健康体检人员肠集聚性大肠埃希菌分子特征及耐药性研究

Antibiotic resistance and molecular characteristics of enteroaggregative Escherichia coli from patients with acute diarrhea and persons receiving health examination

  • 摘要: 目的 比较急性腹泻病患者与健康体检人员肠集聚性大肠埃希菌(EAEC)在携带毒力基因、耐药性及分子分型之间的差异。方法 以PCR方法检测EAEC的7种毒力基因,微量肉汤稀释法检测菌株对13种抗菌药物的耐药性,脉冲场凝胶电泳(PFGE)进行分子分型分析。结果 来自急性腹泻病患者的分离株中,毒力基因检出率最高为air,体检分离株为sat。48株EAEC对13种抗菌药物中的12种均表现出不同程度的耐药,对亚胺培南完全敏感,急性腹泻病患者分离株耐药性较体检分离株强。PFGE分型共得到43种带型,均呈现高度多态性。结论 不同来源EAEC携带的毒力基因差异不显著,PFGE带型呈高度多态性,来源于腹泻病患者的EAEC耐药性明显高于健康体检人员。

     

    Abstract: Objective To compare the differences in the virulence genes, antibiotic resistance and molecular characteristics of enteroaggregative Escherichia coli (EAEC) strains from acute diarrhea patients and persons receiving health examination. Methods The 7 virulence genes were detected by using polymerase chain reaction(PCR), the resistances to 13 antibiotics were tested with micro broth dilution method and pulsed-field gel electrophoresis(PFGE)was conducted for molecular analysis.Results Among the EAEC strains isolated from the diarrhea patients, the detection rate of virulence gene air was highest, and the virulence gene sat was mainly detected in healthy persons. Forty eight EAEC strains were resistant to 12 kinds of antibiotics, but sensitive to imipenem. The drug resistant rate was higher in the strains from diarrhea patients than in the strains from healthy persons. Forty three PFGE patterns were found in EAEC strains with highly polymorphism.Conclusion There was no significant difference in virulence genes between EAEC strains from different sources, and PFGE band was highly polymorphic. The drug resistant rate of EAEC from patients with diarrhea was higher than that from healthy persons.
