江西省赣州市两株冠状病毒的全基因组遗传特征分析

Genetic analysis of complete genome of two coronaviruses strains isolated in Ganzhou, Jiangxi

  • 摘要: 目的 分析江西省赣州市2株亚洲鼩鼱携带冠状病毒的全基因组遗传特征。方法 用反转录-聚合酶链式反应及RACE方法从亚洲鼩鼱粪便中扩增其全基因组;利用MegAlign软件进行序列同源性分析;利用phyML软件构建系统发育树。结果 全基因组的同源性分析表明毒株GZ01、GZ02与已知的亚洲鼩鼱携带冠状病毒的序列具有很高的同源性(88.6%~92.1%)。系统发育分析表明毒株GZ01、GZ02与上述序列有最近的亲缘关系。结论 赣州市亚洲鼩鼱携带属冠状病毒,与已知的毒株为同一种冠状病毒,系统发育分析发现亚洲鼩鼱在冠状病毒的起源与进化过程中起着重要作用。

     

    Abstract: Objective To investigate the genetic characteristics of two strains of coronaviruses carried by Asian house shrew in Ganzhou. Methods The complete genome sequences of 2 coronavirus strains isolated from stool samples of Asian house shrew were amplified by using RT-PCR and RACE. The nucleotide sequences similarity was calculated with software MegAlign. Phylogenetic analysis was performed by using software phyML. Results The nucleotide sequences of whole genome of GZ01 strain and GZ02 strain shared the high homology (88.6%-92.1%) with other strains of coronaviruses previously detected in Asian house shrew. Phylogenetic analysis confirmed that GZ01 and GZ02 strains had the closest relationship with these strains. Conclusion Coronavirus carried by Asian house shrew from Ganzhou belonged to alphacoronaviruses.
