广东省惠州市集中空调系统冷却水及冷凝水中嗜肺军团菌脉冲场凝胶电泳分型分析

Pulsed field gel electrophoresis typing of Legionella pneumophila in cooling water and condensed water from centralized air conditioning system in Huizhou

  • 摘要: 目的 分析广东省惠州市公共场所集中空调系统冷却水及冷凝水中分离的嗜肺军团菌的基因特征,初步建立惠州市嗜肺军团菌脉冲场凝胶电泳(PFGE)分型数据库。方法 采用PFGE技术,对2012-2016年惠州市公共场所集中空调系统冷却水及冷凝水中分离的35株嗜肺军团菌进行电泳分析获得指纹图谱,并用BioNumerics 6.6软件对其进行聚类分析。结果 35株嗜肺军团菌分为29种不同的PFGE带型,菌株间带型相似系数为21.70%~100.00%。每种带型包含1~3株菌,存在5组带型一样的菌株,不存在优势型别;相同PFGE型别的菌株均来源于同一场所;同一场所存在2种以上不同PFGE型别;部分场所的冷却水和冷凝水中分离的菌株存在相同PFGE型别;相同血清型别的菌株经PFGE分型后得到不同的图谱,各菌株间PFGE图谱差异较大。结论 惠州市公共场所集中空调冷却水及冷凝水中分离的嗜肺军团菌呈现基因多样性。

     

    Abstract: Objective To understand the genetic characteristics of Legionella pneumophila isolated from cooling water and condensed water of centralized air conditioning system in the public places of Huizhou, Guangdong province, and establish local L. pneumophila pulsed-field gel electrophoresis (PFGE) typing database. Methods By using PFGE, 35 strains of L. pneumophila isolated from cooling water and condensed water of centralized air conditioning system in the public places of Huizhou from 2012 to 2016 were tested for their fingerprints, and cluster analysis was performed with BioNumerics 6.6 software. Results The 35 strains of L. pneumophila were divided into 29 PFGE types, with the similarity ranging from 21.70% to 100.00%. Each type contained 1-3 strains, and there were 5 groups of strains sharing same type, no predominant PFGE type were found. The strains belonged to the same PFGE type were collected from the same place. There were more than two kinds of different PFGE types in the strains isolated from the same place. Same PFGE type were detected in the strains isolated from cooling water and condensed water collected at same place; The strains belonged to same serotype had different PFGE patterns, indicating that PFGE patterns differed among the strains. Conclusion The strains of L. pneumophila isolated from cooling tower water and condensed water of centralized air conditioning system in the public places of Huizhou showed genetic diversity.
