脉冲场凝胶电泳与质粒图谱分析应用于浙江省甲副伤寒菌株的分子流行病学调查

Application of Pulsed Field Electrophoresis and Plasmid Profile on Molecular Epidemiological Survey of Paratyphoid A Strain in Zhejiang Province

  • 摘要: 目的应用分子生物学方法对浙江省甲副伤寒流行菌株的特征和分子分型进行研究。方法对浙江省26株暴发及散发甲副伤寒菌株作药敏试验、应用快速小量质粒提取方法并作图谱分析,同时应用脉冲场凝胶电泳分型技术(PFGE)分析比较暴发及散发菌株的基因图谱。结果26株菌株对SM、CAR耐药率高于52.00%,对COS、AMP高于84.00%,对SD高达100.00%。所有菌株含有-2.3Kb和-23.1Kb大小质粒。经XbaI酶切,作PFGE分析发现各个菌株的染色体酶切图谱显示19~21条带,26株菌株共出现四种带型,所有菌株分析的条带数仅相差1~2条。结论浙江省甲副伤寒菌株普遍带有质粒且绝大数菌株具多重耐药性,这可能与浙江省甲副伤寒持续性高发有关。PFGE分析所有菌株均为同一克隆系,有一定相关性,说明浙江省流行的甲副伤寒菌株相对保守。

     

    Abstract: Objective This study aimed tO find the characteristics and molecular heterogeneity of the epidemic Paratyphoid A strain in Zhejiang Province through an approach of molecular biol-ogy.Methods A drug sensitive test for 26 fulminant and sporadic strains of Paratyphoid A in Zhejiang Province was conducted,a small amount of plasmid was extracted through a rapid method and a profile analysis was done,and at the same time,the gene maps of fulminant and sporadic strains were analyzed and compared through pulsed field gel electrophoresis (PEGE) technology.Results The drug resistance rates of the 26 strains to SM and CAR are higher than 52.00%.tO COS and AMP higher than 84.00%,and to SD higher than 100.00%.All the strains contain plasmid of-2.3Kb and-23.1Kb.After XbaI enzyme cutting,PFGE analysis was done,and the Macrorestricfion Map of chromosome for all strains showed 19 to 21 strips,and there are four types of strips for the 26 strains.The difference of strips numbers between any two of the strains is only 1-2.Conclusion Generally,the strains of Paratyphoid A in Zhejiang Province have plasmid,and most of the strains are of multiple drug resistance.Maybe this is relevant to the durative high incidence of Paratyphoid A in Zhejiang Province.PFGE analysis found that all strains were of the same clone family and relevant to some extent.This showed that the epidemic strains of Paratyphoid A in Zhejiang Province are conservative relatively.
