钩端螺旋体病MAT检测方法的标准化及结果准确性国际评测

The Standardization of MAT for the Detection of Leptospirosis Antibody and International Testing of theVeracity for MAT Results

  • 摘要: 目的对经典的钩端螺旋体病MAT(显微镜凝集试验)方法进行标准化,以提高其检测结果的可靠性、便于今后不同实验室之间MAT检测结果的对比分析。方法参照世界卫生组织/国际钩端螺旋体病协会(WHO/ILS)新近推荐的MAT操作步骤,对我国MAT实验中的各个环节,包括培养基制备、菌体浓度、抗原制备、血清灭活方法、抗原抗体作用时间与温度,以及结果判定标准等进行标准化。结果用标准化MAT方法检测WHO/ILS寄送的考核盲样(编号A、B、C、D、E),每份样本重复检测三次,获得的实验结果完全一致。检测结果报送WHO/ILS后,反馈结果表明:5份样本检测的血清群结果完全准确,抗体滴度与标准结果吻合。结论钩端螺旋体病MAT实验经标准化后,检测结果稳定可靠,在国际性评测中能准确地鉴定出钩端螺旋体血清抗体的血清群及滴度。标准化方法的应用可提高我国钩体病血清学监测水平。

     

    Abstract: Objective The present study was designed to standardize the traditional MAT methods used for the detection of leptospirosis antibody for the improvement of the reliability of test results,allowing the comparision and analysis of the MAT results obtained in different laboratories.Methods On the base of the methods newly recommended by WHO/ILS,The procedures involved in MAT currently implemented in China,including the preparation of culture medium、leptospira concentration、antigen preparation、serum inactivating,the time and temperature for antigen-antibody reaction and the criterions for the evaluation of result etc.Were standardized.Results Each of the blind samples (encoded as A,B,C,D and E) provided by WHO/ILS was repeatedly detected for three times and the results were the same.And the results were reported to WHO/ILS for confirmation and it was reported that the detected serogroup results of the five samples were accurate,the antibody titres consistent with the standards.Conclusions The standardized procedure of MAT for the detection of leptospirosis allows for more steady and reliable results,testing the exact serogroup and titre of serum leptospirosis antibody perfectly in the international MAT scheme.Standardized MAT contributes the improvement of the serological surveillance of leptospirosis in our country and is worthy of application.
