Identification of A HIV-1 CRF15_01B subtype similar strain in Beijing

  • 摘要: 目的 通过对一例HIV-1毒株env、gag和pol基因的序列分析,表明HIV-1 CRF15_01B亚型类似毒株已在北京市出现.方法 从一例在北京居住的四川籍女性HIV-l感染者(BJ06108,通过性途径感染)血浆中提取病毒RNA,使用套式PCR方法扩增env、gag和pol基因,并进行序列测定和亚型分析.结果 病例BJ06108在env、gag和pol基因区与CRF15_01B亚型参考株1501B.TH.99.99TH_MU2079基因离散率较近,基因离散率分别为8.8%、6.1%和7.2%.系统进化树分析表明BJ06108与CRF15_01B亚型参考株1501B.TH.99.99TH_MU2079聚在一起.结论 CRF15_01B亚型类似毒株已在北京市出现,应该加强HIV-1毒株亚型变异的监测.

     

    Abstract: Objective The study revealed the appearance of an HIV-1 CRF15_01B subtype similar strain in Beijing through sequence analysis of env,gag and pol genes of the strain.Methods The plasma viral RNA was extracted from a HIV-1 infected(BJ06108,through sexual transmission) Sichuan woman living in Beijing.Nested PCR was used to amplify the env,gag and pol genes. Sequencing and subtype analysis was then performed.Results The discrete rates in env,gag and pol gene regions of case BJ06108 were similar to those of the CRF15_01B subtype reference strain,1501 B.TH.99.99TH_MU2079 being 8.8%,6.1% and 7.2%,respectively.Phylogenetic tree analysis showed that BJ06108 and the CRF15_01B subtype reference strain,1501 B.TH.99.99TH_MU2079,shared a same node.Conclusion A CRF15_01B subtype similar strains had appeared in Beijing.It was necessary to strengthen the surveillance of HIV-1 subtype variation.
