鉴别29E群、X群和Z群脑膜炎奈瑟菌聚合酶链反应方法的建立及应用

Establishment and application of PCR assay for identification of <I>Neisseria meningitidis </I>serogroup 29E, X, and Z

  • 摘要: 目的建立鉴别29E群、X群和Z群脑膜炎奈瑟菌的聚合酶链反应(PCR)方法。方法针对脑膜炎奈瑟菌IctrA/I基因变异区序列,设计合成PCR引物,分别用于扩增29E群、X群和Z群等3个血清群脑膜炎奈瑟菌。并将该方法应用于41份疑似脑膜炎奈瑟菌菌株标本的检测。结果3对引物分别能够特异性PCR扩增29E群、X群和Z群脑膜炎奈瑟菌菌株的IctrA/I基因,扩增片段大小分别为667 bp、525 bp和667 bp,3个血清群之间以及与其他血清群脑膜炎奈瑟菌之间未出现交叉PCR扩增。41份疑似脑膜炎奈瑟菌菌株标本中,29E群和X群PCR阳性的标本分别有7份和2份,未检测出Z群菌株。结论本研究建立的PCR方法能准确、特异地鉴定29E群、X群和Z群脑膜炎奈瑟菌菌株。

     

    Abstract: ObjectiveTo establish PCR assay to identify INeisseria meningitides/I serogroup 29E, X and Z. MethodsPCR primers targeting the variation regions of IctrA/I gene were designed and synthesized to amplify IN.meningitidis/I sewrogroup 29E, X and Z. The established PCR assay was used to detect 41 samples of suspected IN.meningitidis/I isolates. ResultsThe IctrA/I gene from IN.meningitidis/I serogroup 29E, X and Z could be amplified with the three pairs of primers respectively. The amplicons were 667 bp (29E), 525 bp (X), and 667 bp (Z). No cross-reactions were observed among three serogroups or with other serogroups. Of 41 samples isolated, 7 were positive for serogroup 29E and 2 for serogroup X. ConclusionThe PCR assay in this study could indentify N. meningitidis serogroup 29E, X, and Z accurately and specifically.
