万古霉素耐药肠球菌的药敏表型及基因特性研究

Susceptibility phenotypes and genetic characteristics of vancomycin-resistant Enterococci

  • 摘要: 目的 研究万古霉素耐药肠球菌(VRE)的耐药表型和基因特性。 方法 采用浓度梯度法(E-test)测定VRE对万古霉素(VA)、替考拉宁(TP)的耐药表型;采用聚合酶链反应检测vanA、vanB、vanC1、vanC2、vanC3基因,并对van基因产物进行测序。 结果 万古霉素耐药基因检测结果:VRE经PCR扩增,均获得vanA基因阳性片段。未检测到vanB、vanCl及vanC2/3基因。 结论 万古霉素耐药肠球菌的耐药表型与耐药基因型一致,菌株之间有较高的同源性。住院患者肠道中VRE携带率高,是医院感染的危险因素。

     

    Abstract: Objective To understand the susceptibility phenotype and genetic characteristics of vancomycin-resistant Enterococci (VRE). Methods Epsilometer test was conducted to detect the resistance of VRE to vancomycin and teicoplanin. PCR was conducted to detect vanA, vanB, vanC1, vanC2 and vanC3 gens, and related PCR products were confirmed by DNA sequencing. Results The result of vancomycin resistant gene detection indicated that all the VRE strains had vanA gene, no vanB, vanCl and vanC2/3 gens were detected. Conclusion Susceptibility phenotypes of VRE were consistent with their vancomycin resistant genotypes. The strains shared high homology. VRE carriage is high in intestinal tract of inpatients, which is a risk factor for nosocomial infection.
