2008-2013年浙江省柯萨奇病毒A组16型VP1区基因特征分析

Genetic characteristics of VP1 region of Coxsackie A16 virus in Zhejiang, 2008-2013

  • 摘要: 目的 了解浙江省2008-2013年手足口病(hand foot and mouth disease, HFMD)患者中柯萨奇病毒A组16型(Coxsackievirus A16,Cox A16)的流行株VP1区基因特征. 方法 对浙江省2008-2013年手足口病患者粪便、疱疹液和咽拭子标本中分离到的Cox A16毒株用反转录-聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)进行VP1区特异性基因扩增,对扩增产物进行核苷酸序列测定并参考Cox A16的参考毒株和中国其他地区的Cox A16分离毒株进行同源性分析和构建系统发生树. 结果 浙江省2008-2013年的81株Cox A16分离株,其VP1区核苷酸和氨基酸同源性分别是88.7%~100%和97.8%~100%.与原型株G10(G-10/RSA/1951)的VP1区核苷酸和氨基酸同源性分别是61.9%~65.1%和90.1%~91.7%.81株Cox A16分离株全部属于B1基因亚型,有B1a和B1b两个基因亚型. 结论 浙江地区2008-2013年分离到的81株Cox A16分离株属于B1基因亚型,有B1a和B1b两个进化分支共同进化和循环.

     

    Abstract: Objective To understand the genetic characteristics of VP1 region of Coxsackie A16 virus(Cox A16) isolated from hand foot and mouth disease(HFMD) patients in Zhejiang province during 2008-2013. Methods Reverse transcription-polymerase chain reaction(RT-PCR) was conduct to amplify the VP1 region genes of Cox A16 isolated from clinical specimens(stool, herpes fluid and throat swab). Homologous comparison and phylogenetic analysis of Cox A16 strains were performed by using reference Cox A16 strain and Cox A16 strains isolated from other areas of China. Results A total of 81 Cox A16 strains were isolated, and they shared 88.7%-100% homology of nucleotide and 97.8%-100% homology of amino acid of VP1 region. Compared with the prototype strain of Cox A16(G-10/RSA/1951), the homologies of nucleotide and amino acid were 61.9%-65.1% and 90.1%-91.7%, respectively. The phylogenetic tree indicated that all the 81 Cox A16 strains belonged to gene subtype B1(B1a and B1b). Conclusion The Cox A16 strains isolated in Zhejiang from 208 to 2013 belonged to gene subtype B1 and could be further classified as B1a and B1b.
