宁波地区金黄色葡萄球菌临床株肠毒素基因与多位点序列分型研究

Enterotoxin genes detection and multi-locus sequence typing of Staphylococcus aureus isolated from patients in Ningbo

  • 摘要: 目的 了解宁波地区金黄色葡萄球菌(Staphylococcus aureus)临床株肠毒素基因分布和多位点序列分型特征。方法 收集宁波市各医院分离的金黄色葡萄球菌临床株,利用聚合酶链反应(polymerase chain reaction,PCR)检测金黄色葡萄球菌肠毒素A(sea)、金黄色葡萄球菌肠毒素B(seb)、金黄色葡萄球菌肠毒素C(sec)和金黄色葡萄球菌肠毒素D基因(sed),对肠毒素基因阳性菌株利用多位点序列分型(multi-locus sequence typing, MLST)进行分子分型。结果 2006、2008和2011-2013年共收集临床株155株,其中肠毒素基因阳性株为45株,阳性率为29.03%;4种基因阳性率依次为16.77%、5.81%、11.61%和3.87%;形成8种基因组合,以sea(14株)、sec(9株)和sea+sec (8株)三种组合为主。45株菌株可分为15个ST(sequence type,ST),ST1有14株,占31.11%;ST5有6株,占13.33%; ST1121和ST15的菌株均为4株,各占8.89%。与全国其他地区比较,在宁波地区临床株中发现ST12、ST573和ST1121三种新的ST。45株菌株在系统进化树上形成7个主要分支。在ST1克隆群中,sea阳性率为72.73%(16株),sec阳性率为54.55%(12株),以sea+sec组合为主(8株)。结论 宁波地区金黄色葡萄球菌临床株肠毒素基因阳性率较高,以携带sea和sec为主, 形成8种基因组合。有15种ST型,发现ST12、ST573和ST1121三种新的ST。ST1克隆群为流行优势菌株,其次为ST5克隆群。ST1克隆群以携带sea和sec为主。

     

    Abstract: Objective To investigate the enterotoxin genes distribution and molecular characteristics of Staphylococcus aureus isolated from patients in Ningbo. Methods S. aureus strains were collected from the patients. S. aureus enterotoxin gene A(sea), S. aureus enterotoxin gene B(seb), S. aureus enterotoxin gene C(sec)and S. aureus enterotoxin gene D(sed)were detected with polymerase chain reaction (PCR). Molecular subtyping were conducted with multi-locus sequence typing (MLST). Results A total 155 clinical strains were collected in 2006,2008 and during 2011-2013, in which 45 were enterotoxin gene positive. The positive rates of 4 enterotoxin genes were 16.77%, 5.81%, 11.61% and 3.87% respectively. The 45 strains formed 8 kinds of assortment of genes. Sea (14 strains), sec (9 strains) and the combination of sea and sec (8 strains) were predominant. The 45 strains could be divided into 15 sequence types (STs) with MLST. ST12, ST573 and ST1121 were newly found in Ningbo compared with other areas of China. ST1 (14 strains), ST5 (6 strains), ST1121 (4 strains) and ST15 (4 strains) were the major STs. The positive rate of 4 STs was 62.22%. The phylogenetic tree had 7 branches. The positive rate of sea and sec genes were 72.73% and 54.55% in ST1 clone complex. The combination of sea and sec was the major genes in ST1 clone. Conclusion The positive rate of enterotoxin gene was high in S. aureus isolated from the patients in Ningbo. Most of the clinical strains carried sea and sec genes. There were 15 STs circulating in Ningbo. ST12, ST573 and ST1121 were newly found in clinical strains isolated in Ningbo. ST1 clone complex was the predomiant genotype, followed by ST5 clone complex. Most of the ST1 clone complex carried sea and sec genes.
