结核分枝杆菌中假设蛋白抗原表位的多态性研究

Polymorphism of human T cell epitopes of eight immune-related conserved hypothetical proteins in Mycobacterium tuberculosis

  • 摘要: 目的 探究中国结核分枝杆菌(Mycobacterium tuberculosis)菌株中8个免疫相关的保守性假设蛋白Rv1158c、Rv1461、Rv1977、Rv2182c、Rv3207c、Rv3333c、Rv3378c和Rv3714c人T细胞表位的多态性。方法 选取151株我国临床分离到的结核分枝杆菌菌株,对8种抗原基因进行PCR扩增,并运用Bioedit软件进行序列比对,比较其人T细胞表位的多态性。结果 在151株菌株中,除了Rv1977外其余7个蛋白都表现保守,这与Comas等的研究结果一致。而Rv1977表现了一定的多态性,发生了3个异义突变和一个单碱基的缺失。在Rv1977中的3个T细胞抗原表位中有1个表位发生了变化。结论 结核分枝杆菌中的8个与免疫相关的保守性假设蛋白的T细胞抗原表位中,大部分是保守的,只有Rv1977蛋白具有多态性,这种多态性可能反映了这个抗原参与了逃避宿主免疫的分化选择。

     

    Abstract: Objective To investigate the polymorphisms of human T epitopes of antigens Rv1158c, Rv1461, Rv1977, Rv2182c, Rv3207c, Rv-3333c, Rv3378c and Rv3714c in Mycobacterium (M.) tuberculosis isolated in China. Methods We selected 151 clinical isolates of M. tuberculosis for the amplification of gene sequences of the eight antigens. The sequence alignment was conducted with software Bioedit. Results Among the 151M. tuberculosis strains, all the proteins were conservative in T cell epitopes except Rv1977. There were three non-synonymous mutations and one frame shift mutation in Rv1977. The mutations and deletion in Rv1977 all affected one of three T cell epitopes. Conclusion Among 8 immune-related conserved hypotheticals proteins in M. tuberculosis, only Rv1977 showed polymorphism in T cell epitopes, indicating that the antigen might be involved in diversifying selection to evade host immunity.
