secA1基因用于诺卡菌菌种鉴定分型的研究

Identification of Nocardia species based on secA1 gene

  • 摘要: 目的 研究secA1基因在诺卡菌菌种的鉴定分型能力。方法 对59株诺卡菌的secA1基因进行聚合酶链反应扩增,扩增片段纯化后测序,并从GenBank数据库下载32株诺卡菌secA1基因序列进行补充,采用DNAStar软件对所有序列计算种内与种间相似度水平,并用Mega 6.06软件的邻接法构建诺卡菌菌种系统发育进化树。结果 91株受试诺卡菌secA1基因的种内相似度为97.2%~100.0%,平均相似度达98.6%;种间相似度为85.2%~98.4%,平均相似度达91.8%。以secA1基因构建的系统发育进化树中,16种诺卡菌被准确分离为16个独立的分支,诺卡菌近缘种新星诺卡菌复合体、少食/短链诺卡菌复合体均可有效分离。结论 secA1基因序列分析为诺卡菌菌种的鉴定分型提供了一种新方法,在诺卡菌近缘种的分型方面有独特的优势。

     

    Abstract: Objective To evaluate the role of secA1 gene for identifying and typing Nocardia species. Methods secA1 gene sequences of 59 Nocardia strains were amplified by polymerase chain reaction (PCR) and purified products were sequenced and then used for similarity analysis of secA1 by using DNAStar software, meanwhile the secA1 gene sequences of another 32 Nocardia strains were downloaded from GenBank. The phylogenetic tree based on secA1 gene was constructed by using Mega 6.06 software (neighbor-joining method). Results The intra and inter species similarity of 91 strains of Nocardia was 97.2% to 100.0% and 85.2% to 98.4% respectively (averagely 98.6% and 91.8% respectively). In the phylogenetic tree based on secA1 gene sequences, the 16 Nocardia species were accurately divided into 16 different clusters, especially the close species, including Nocardia complex. Conclusion Sequencing of secA1 is a novel method for identifying and typing Nocardia species, especially for the close species.
