我国羊种布鲁氏菌rpoB基因遗传多态性分析

Genetic polymorphism analysis of rpoB gene of Brucella melitensis strains isolated in China

  • 摘要: 目的 调查中国羊种布鲁氏菌rpoB基因的多态性。方法 利用AMOS-PCR对分离菌株进行种型鉴定;根据布鲁氏菌rpoB基因的4个多态性位点设计特异性引物,对299株羊种布鲁氏菌流行株(羊1型菌38株、羊2型菌19株、羊3型菌242株)的rpoB基因的4个多态性位点进行扩增,并将PCR产物测序结果与羊种布鲁氏菌标准菌株的扩增结果进行比对。结果 羊种布鲁氏菌的rpoB基因检测发现,有169株为rpoB-2变异型、103株为rpoB-2型、24株为新的rpoB基因型(rpoB-4型)及3株为rpoB-1型。结论 我国羊种布鲁氏菌rpoB基因具有较高的多态性,可作为羊种布鲁氏菌的分子流行病学调查靶基因,我国羊种菌rpoB基因型和生物型无相关性。

     

    Abstract: Objective To investigate polymorphism of rpoB gene of Brucella melitensis strains isolated in China. Methods Species/biovars of B.melitensis stranis were identified by AMOS-PCR. Then,specific primers were designed according to four polymorphic loci of Brucella rpoB gene,four diversity loci of rpoB gene of 299 B.melitensis stranisincluding B.melitensis biovar 1(38),B.melitensis biovar 2(19),B.melitensis biovar 3(242)were amplified,and sequencing results of PCR product were compared with rpoB gene of standard strains of B.melitensis. Results Analysis of rpoB gene of 299 B.melitensis strains revealed that 169 strains were rpoB-2 variants genotype,103 were rpoB-2 genotype,24 strains exhibited new rpoB genotype(named as rpoB-4),3 strains of rpoB-1 genotype. Conclusion RpoB gene of B.melitensis from China has higher genetic polymorphism,rpoB gene can be used as a target gene for molecular epidemiological investigation of B.melitensis.
