浙江省衢州市首例输入性基孔肯雅热病毒基因特征分析

Genetic characteristics of Chikungunya virus isolated from the first imported Chikungunya fever case in Quzhou, Zhejiang

  • 摘要: 目的对1例有孟加拉国旅行史的基孔肯雅热病例进行病毒基因特征分析。方法采集患者血清,采用实时荧光定量PCR(real-time PCR)检测基孔肯雅热病毒(CHIKV)和登革热病毒核酸,反转录PCR(RT-PCR)扩增CHIKV结构基因C-E3-E2-6K-E1片段并测序,计算机软件分析处理基因序列。结果患者血清real-time PCR检测为CHIKV核酸阳性,病毒株编号为QZ0823。 对病毒株QZ0823病毒进行测序拼接后共获得CHIKV 5个结构基因全长,包含3 747 bp核苷酸序列,编码1 248个氨基酸,与参考序列核苷酸和氨基酸同源性分别为98.9% ~ 99.9%和99.0% ~ 99.9%,对E1和C-E3-E2-6K-E1核苷酸进行系统进化分析,病毒株QZ0823株与孟加拉国、印度、巴基斯坦的东中南非洲遗传谱系流行株进化关系最近。结论来自病例的CHIKV病毒株QZ0823属于东中南非洲遗传谱系,该病例为输入性病例。

     

    Abstract: ObjectiveTo understand the genetic characteristics of Chikungunya virus isolated from the first imported Chikungunya fever case with travel history in Bangladesh in Quzhou, Zhejiang province.MethodsThe serum sample was collected from the case for the detections of the nucleic acids of CHIKV and Dengue virus with real-time PCR. The gene C-E3-E2-6K-E1 fragment was amplified by using RT-PCR. The PCR products were sequenced and used for tracing analysis by using Bioinformatics software.ResultsThe positive bands of the specific nucleic acids of the CHIKV strain (QZ0823) isolated from the case could be amplified by PCR. Five structural genes of the CHIKV strain were obtained after splicing the sequencing data, including a 3 747 bp nucleotide sequence which encoded 1 248 amino acids. The nucleotide acid and amino acid of the strain shared 98.9%−99.9% and 99.0%−99.9% homologies, respectively with the reference sequences. Phylogenetic analysis of the nucleotide sequences of the E1 and C-E3-E2-6K-E1 genes revealed that the strain had close genetic relationships with the strains from Bangladesh, India, and Pakistan belonging to the genetic pedigree of East Central and South Africa.ConclusionThe CHIKV strain (QZ0823) isolated in Quzhou belonged to genetic pedigree of East Central and South Africa, indicating that the Chikungunya fever case in Quzhou was imported one.
