天津市儿童人博卡病毒流行状况及基因特征分析

Detection and genetic characteristics of human bocavirus in children in Tianjin

    目的了解天津地区急性呼吸道感染儿童中人博卡病毒(HBoV)的感染情况及病毒基因特性和变异情况。
    方法采集2018年5月至2019年4月因急性呼吸道感染住院儿童的呼吸道标本248份,采用多重荧光定量聚合酶链式反应方法检测多种呼吸道病毒,对检测出的HBoV阳性标本和2015 — 2018年本实验室留存的HBoV阳性标本,扩增VP1及VP2基因全序列,并进行序列测定,与GenBank中序列进行比较,利用DNAStar和MEGA软件进行拼接并绘制种系发生树分析。
    结果248份标本中6.05%(15/248)为HBoV阳性,阳性患儿的年龄6个月至4岁,所有感染者均为肺炎或支气管炎,10月和11月阳性检出率最高。 测序成功的17株HBoV均属于HBoV-1型,以其中的Ⅰb簇流行为主,但同时也有Ⅰa簇和其他簇存在,天津地区毒株间以及与标准株ST2和ST1间的核酸同源性非常高,为97.9%~100.0%,蛋白质在影响抗原性的主要区域总共有6处氨基酸发生了变异。
    结论天津地区HBoV流行高峰期为10月和11月,HBoV流行株以Ⅰb簇为主,基因特性较为稳定,但仍有个别关键位点发生改变。

     

    Abstract:
    ObjectiveTo investigate the infection status of human bocavirus (HBoV) in infants and young children with acute respiratory tract infection in Tianjin and identify molecular characteristics of the virus.
    MethodsRespiratory samples were taken from 248 hospitalized pediatric patients in Tianjin from May 2018 to April 2019. The pathogens were detected by using multiplex real-time PCR kit for respiratory viruses. VP1/VP2 genes of the HBoV strainsisolated from the HBoV positive samples and the HBoVstrains stored in our laboratory from 2015 to 2018 were amplified and sequenced.The sequences were analyzed,phylogenetic tree was constructed using software DNAstar and Mega.
    ResultsOf all 248 pediatric patients, 15 (6.05%) were positive for HBoV. The HBoV-positive children were aged 6 months-4 years old and all had pneumonia or asthmatic bronchitis. The detection rate of HBoV peaked during October - November in Tianjin. The VP1/VP2 genes of 17 HBoVstrains were successfully sequenced, and phylogenetic analyses showed that they all belonged to HBoV-1, and Ⅰb cluster was predominant. The nucleic acid identity was 97.9%–100.0% compared with standard strains ST2 and ST1. There were 6 key amino acid mutations within antigenic domains.
    ConclusionThe incidence of HBoV peaked during October-November in Tianjin, most HBoV strains in Tianjin belonged to Ⅰb cluster. The genetic characteristics of HBoVwere stable without obvious variation, but some key amino acid mutations were observed.
