浙江省龙游县鼠形动物中5种病原的自然感染情况调查

Natural infection status of 5 pathogens in rodents in Longyou county, Zhejiang

      目的  了解浙江省衢州市龙游县鼠形动物中5种病原的自然感染情况,为预防和控制相关疾病提供依据。
      方法  在龙游县野外捕获鼠形动物,解剖获取肺和肝脏组织。 对鼠肺进行冷冻切片,用冷丙酮固定切片后用直接免疫荧光法检测汉坦病毒的携带情况;提取鼠肝DNA,利用特异性引物进行PCR扩增,检测伯氏螺旋体、查菲埃立克体、无形体、鼠疫耶尔森菌。
      结果  2017年捕获的黑线姬鼠、针毛鼠、社鼠、臭鼩鼱、黄胸鼠和褐家鼠(共6种88只)中,黑线姬鼠为优势鼠种(88.64%),其次为针毛鼠(5.00%),其他种数量很少。 2019年捕获的黑线姬鼠、黄胸鼠、针毛鼠、褐家鼠、臭鼩鼱和巢鼠中(共6种103只),黑线姬鼠也为优势鼠种(84.47%)。 病原检测结果发现,在2017、2019年捕获的黑线姬鼠中,汉坦病毒抗原的阳性率分别为3.85%和4.60%,伯氏螺旋体核酸阳性率分别为1.28%和2.30%,查菲埃立克体核酸阳性率分别为6.41%和5.75%,无形体的核酸阳性率分别为1.28%和2.30%,鼠疫耶尔森菌均阴性。 其余鼠种均未检出相关病原。
      结论  龙游县黑线姬鼠数量多,是多种重要传染病的潜在传染源,需重点监测,扩大相关病原谱的调查和监测,为自然疫源性疾病的预防控制提供依据。

     

    Abstract:
      Objective  To investigate the natural infection status of 5 pathogens in rodents in Longyou County of Zhejiang province, and provide evidence for the prevention and control of related diseases.
      Methods  Rodents were captured in the field in Longyou. The lungs of the captured rodents were frozen and fixed in cold acetone. Hanta virus was detected by direct immunofluorescence. Rodent liver DNA extraction was done for the detection of Borrelia burgdorferis, Ehrlichia chaffeensis, Phagocytophilum, and Yersinia pestis by PCR amplification.
      Results  A total of 88 rodents in 6 species were captured in 2017, including Apodemus agrarius, Rattus acutus, Rattus commune, shrew, Rattus flavipectus and Rattus norvegicus. Among them, Apodemus agrarius was the predominant species (88.64%), followed by Rattus acutus (5.00%). In 2019, a total of 103 rodents in 6 species were captured, including Apodemus agrarius, Rattus flavipectus, Rattus acutus, Rattus norvegicus, shrew and nested rat. Apodemus agrarius was still the predominant species, accounting for 84.47% of the total captured rodents. The results of pathogen detection showed that the positive rate of Hanta virus antigen was 3.85% and 4.60% in 2017 and 2019, respectively. The positive rate of Borrelia burgdorferi nucleic acid were 1.28% and 2.30%, respectively. The positive rate of Ehrlichia chaffeensis nucleic acid was 6.41% and 5.75%, respectively. The positive rate of Phagocytophilum nucleic acid was 1.28% and 2.30%, respectively. The detection results of Yersinia pestis were negative. No related pathogens in other species were detected.
      Conclusion  There is a large number of Apodemus agrarius in Longyou, which is a potential infection source of many important infectious diseases. Therefore, it is necessary to strengthen the surveillance for Apodemus agrarius. In addition, the investigation of related diseases should be expanded in the future to provide evidence for the prevention and control of natural focal diseases.
