创伤弧菌脓毒症大鼠肝细胞Bcl-2 mRNA、Bax mRNA表达的动态变化研究

Changes on gene expression of Bcl-2 and Bax mRNA in hepatocyte of rats with sepsis of Vibrio vulnificus

摘要: 目的探讨大鼠创伤弧菌(VV) 脓毒症时肝细胞Bcl-2 mRNA、Bax mRNA表达量的动态变化情况。方法制作大鼠VV脓毒症模型(VV组), RT-PCR法测定染菌后各时间点大鼠肝细胞Bcl-2 mRNA、Bax mRNA表达量。结果感染创伤弧菌后大鼠肝细胞Bcl-2 mRNA表达量明显下降(P﹤0.05);Bax mRNA其表达量明显增加(P﹤0.05),染菌后2 h明显增高,在染菌16 h的表达量仍明显高于正常水平(P﹤0.05)。结论创伤弧菌脓毒症可致肝组织中Bcl-2 mRNA表达量下降,Bax mRNA表达量增加,早期明显增高,并持续维持在高水平。

Abstract: Objective To understand the changes on gene expression of Bcl-2 mRNA and Bax mRNA in hepatocyte of rats with sepsis of Vibrio vulnificus. Method Vibrio vulnificus sepsis rats model (VV groups) was established and Bcl-2 mRNA and Bax mRNA were detected by using RT-PCR. Results The gene of Bcl-2 can be detected in hepatocyte of normal rats. Compared with the NC group, the expression of Bcl-2 in VV groups decreased obviously (P﹤0.05). There was Bax mRNA expression in hepatocyte of normal rats. Compared with the NC group, the expression of Bax mRNA increased obviously in VV groups (P﹤0.05), and the gene expression level remained stable Conclusion This study suggested that the expression of Bax mRNA in hepatocyte of rats reached peak in the early period of VV sepsis, while the expression of Bcl-2 mRNA decreased.
