2009年宁波市甲3亚型流感流行特征 与HA1基因特性的分析

Genetic characteristics of HA1 of influenza A (H3N2) virus circulating in Ningbo in 2009

  • 摘要: 目的 了解2009年宁波市甲3亚型流感病毒的流行特征及血凝素基因HA1区域的变异情况。 方法 全年度每周从流感监测哨点医院收集流感样病例标本进行病毒分离,获得的甲3亚型流感毒株按不同时间、地点进行血凝素基因HA1区核苷酸序列测定并推导出其氨基酸序列,然后进行基因特性分析。 结果 所选取的15株甲3亚型流感病毒HA1区域核苷酸序列长度均为987 bp,编码329个氨基酸;2009年上半年的流行株与2008年秋季的流行株在该区域有7个氨基酸位点存在差异,2009年下半年的流行株与2009上半年又存在10个氨基酸位点的差异。上半年与下半年的流行株在种系发生树上形成两个分支,为不同性状的流行株。 结论 2009年上半年及下半年宁波市甲3亚型流感流行株与2008年相比,在血凝素基因HA1区域都发生了较为明显的改变,且上半年与下半年的2个流行株之间血凝素基因的差异也较大。说明2009年宁波市流行的甲3亚型流感病毒变异较为频繁。

     

    Abstract: Objective To understand the circulation of influenza A (H3N2) virus and the antigen variation of the HAl domain of the virus in Ningbo in 2009. Methods The virus was isolated from the influenza like illness cases in sentinel hospitals every week during the year, the nucleotide sequencing of HA1 was conducted for H3N2 isolates and their amino acid sequences were deducted to conduct genetic characteristics analysis. Results The full length of HAl of 15 H3N2 strains was 987 bp, coding protein of 329 amino acids. Compared with the circulating strain in the autumn of 2008, 7 amino acids loci were different in the first half of 2009, and 10 amino acids loci were different between the circulating strain in the first half and that in the second half of 2009. The strains isolated in the first half and in the second half of 2009 belonged to different clades. Conclusion Firstly, compared with the strains isolated in 2008, there were several obvious variations in the HAl domain of strains isolated in the first half and in the second half of 2009; secondly, there were distinct differences between the strains isolated in the first half and in the second half of 2009, thirdly, the influenza A (H3N2) virus varied frequently during 2009 in Ningbo
