32P与光敏生物素标记DNA探针在疟疾监测中的应用研究

Studies on the Application of α—p—Datp of Bio—11—Dutp—labelled Pbf DNA Probe in the Surveilolace of Malaria

  • 摘要: 本文报告了应用32P和光敏生物素标记克隆的pBF4DNA,作为探针,以斑点杂交试验,检测海南、云南和山东省的血样。两种标记探检测结果与镜检的符合率分别为恶性疟98.6%和96.2%;间日疟91.6%和86.7%;正常人血95.2%和100%;并可从多种疟原虫标本中,特异地检出恶性疟原虫,显示探针具有恶生疟原虫的种特异性。检测的敏感度为84个原虫/μl血,检测原虫DNA可达10pg水平。表明探针具有良好的实用性。

     

    Abstract: This report deals with cloned Pbf DNA probe from the genomic DNA library of plas-modium falaiparum labelled with-p-Datp of BIO-11-Dutp was used in dot hybridization to detect blood samples from Hainan,Yunnan and Shandong.The results have shown the coincidence rate of de-tection between those probe and with microscope respectively were 98.6%,96.2%for Plasmodium falciparum;91.6%,86.7% for Plasmodium vivax and 95.2%,100% for normal controls.Plasm modium falciparum could be identified specifically from samples with various malaria parasites,indi-cating that these probe have high specificity for falciparum maolaria infecton.The parasites as low as 84ul blood or 10 pg of parasite DNA could still be discriminated.
