浙江省义乌市一例输入性登革热的病毒分离及全基因组序列分析

Complete genome sequence analysis of Dengue virus 1 isolated in Yiwu, Zhejiang

  • 摘要: 目的 调查浙江省义乌市2013年1例输入性登革热病例的分子流行病学特点。 方法 采集患者血清标本,采用酯酶联免疫吸附试验和实时荧光定量聚合酶链反应分别检测登革病毒IgM抗体和核酸。用C6/36细胞分离病毒和全基因组测序及分析。 结果 从来自安哥拉的1例登革热患者血清中检测到登革病毒IgM抗体和1型登革病毒核酸,并分离到1株病毒(Zj/yw01)。经序列测定,该病毒基因组全长10 141 bp(GenBank no. KF864667),进化树显示该病毒为登革1型病毒Ⅴ亚型,与安哥拉分离株(KF184957)同源性最高。 结论 分子生物学证实该病例为来自安哥拉的输入性病例。

     

    Abstract: Objective To understand the epidemiologic characteristics of an imported dengue fever case in Yiwu in 2013. Methods Blood samples were collected from the case to detect IgM antibody and nucleic acid of the virus with ELISA and real-time RT-PCR and isolate the virus strain with C6/36 cell. The complete genome of the virus was sequenced and analyzed. Results IgM and RNA of dengue virus 1 were detected in the serum sample, the Dengue virus were isolated and named Zj/yw01. The Zj/yw01 strain reaches 10 141 bp in lengthen of complete genome, the viral genomic sequence dada was submitted to the GenBank (Accession No. KF864667). The strain belonged to Dengue virus 1 subtype Ⅴ, which shared highest homology with the virus strain isolated in Angola (KF184957). Conclusion Bio-molecular analysis indicated that the imported dengue fever case was probably originated form Angola.
