Enterohemorrhagic Escherichia coli induced IL-1β level in human macrophages

  • 摘要: 目的 通过构建EHEC O157:H7野生株和突变株等不同菌株,来感染THP-1细胞,从而比较以上不同菌株的细胞毒和IL-1β等细胞因子的产生水平,同时比较使用有无胎牛血清(FBS)培养的THP-1对产生细胞因子水平的影响。 方法 将上述各菌株分别定量稀释于RPMI1640,5%FBS的细胞培养液中,在感染后2~10 h,用cytotox96试剂盒定量检测乳酸脱氢酶(LDH)的释放。同时用Luminex和酶联免疫吸附试验试剂盒定量检测多种细胞因子的水平。 结果 EHEC -O157:H7-ehxA是THP-1细胞毒作用的主要贡献者,并可引起高水平IL-1β的产生;在OPT1-MEM无血清培养基中,野生型菌株和突变型之间IL-1β的产生差异则更加显著。 结论 EHEC-O157:H7-ehxA 可诱导高水平IL-1β的产生,FBS能刺激细胞产生一定非特异性的IL-1β,这可能会导致高的背景和隐藏细胞株之间的真正差别。

     

    Abstract: Objective In this study, we constructed different strains of EHEC O157: H7 (wild strain and mutant strains) to infect THP-1 cells in order to study the toxicity of different strains and cytokine production levels, such as IL-1β, and also compare the THP-1-secreted cytokine levels under the conditions of being cultured with or without FBS after infected with EHEC. Methods Aliquots of the bacteria were added in triplicate to the cell monolayer. At 2-10 h after infection, the release of lactate dehydrogenase (LDH) was quantified by using a Cytotox96 Kit. Meanwhile, the levels of human cytokines were quantified by using a Luminex Kit and ELISA Kit. Results Our results demonstrated that EHEC O157:H7-ehxA contributed to cytotoxicity in THP-1 cells, causing the production of higher IL-1β. Most important, the results showed that the cells cultured in 5% FBS-containing RPMI 1640 medium produced higher IL-1β while the difference in IL-1β level between wild type strains and mutant strains was more significant in OPT1-MEM serum-free medium. Conclusion ehxA plays a crucial role in EHEC O157:H7-induced IL-1β production. Moreover, the result revealed that the serum could stimulate cells to produce small amount of non specific IL-1β, which might cause the high background and hide the real difference between strains.
