Enteric bacteria isolation from plateau pika with two media at the gas condition of 5% CO2

  • Objective To understand whether the new species of Streptococcus exists in the intestinal contents of plateau pika with brain heart infusion medium and KF medium. Methods With two media,i.e. heart infusion medium containing 5% defibrinated sheep blood and KF medium containing 5% defibrinated sheep blood,and one gas condition,i.e. 5% CO2,microorganism was isolated from intestinal contents of plateau pika. The identification was done by using full length of 16S rDNA. Results A total of 607 strains were isolated,which were identified into 19 genera and 31 species. And 495 strains in 19 genera were identified by using the medium of brain heart infusion containing 5% defibrinated sheep blood,and 112 strains in 3 genera were identified by using KF medium containing 5% defibrinated sheep blood.At the level of genus,Arthrobacter,Enterococcus,Bacillus were predominant,accounting for 31.47%,16.14% and 15.98% respectively. The most abundant species were Arthrobacter luteolus,Enterococcus faecium. Some identified species were with medical significance,including Yersinia enterocolitica,Staphylococcus haemolyticus,Staphylococcus epidermidis. Conclusion Bacteria in abundant genera were isolated from the intestinal contents of plateau pika with brain heart infusion medium with 5% defibrinated sheep blood,and the new species of Streptococcus was not found in this experiment. The plateau pika in Qinghai-Tibet area carry some bacterial pathogens.
