副溶血弧菌O10:K4血清型O抗原基因簇的序列特征与分子血清型数据库构建

Sequence characteristics of O-antigen gene cluster of Vibrio parahaemolyticus O10:K4 and construction of molecular serotype database

    目的 分析副溶血弧菌O10:K4血清型O抗原基因簇的分子特征,并构建基于基因组的分子血清型预测数据库。
    方法 从美国国家生物技术信息中心Assembly数据库获取副溶血弧菌O抗原基因簇序列,使用blastn、fastANI、Sourmash和pixy工具分析序列相似性和多态性,使用gubbins进行重组分析,并构建了副溶血弧菌分子血清型预测数据库。
    结果 O10:K4的O抗原基因簇(OL10V1)与传统O10血清型基因簇(OL10)序列差异显著,而与OL4最为相似。 公共数据库中116个OL10V1全长序列仅存在1个单核苷酸突变。 OL10V1携带的IS1634ISVa2转座酶基因广泛分布于弧菌属,提示该变异体可能通过水平基因转移形成。
    结论 本研究揭示了O10:K4血清型快速传播的潜在分子机制,构建的分子血清型数据库可为基因组监测和溯源提供关键技术支撑。

     

    Abstract:
    Objective  To analyze the molecular characteristics of the O-antigen gene cluster of Vibrio parahaemolyticus O10:K4 and construct a genomic-based molecular serotype predictive database.
    Methods The O-antigen gene cluster locus (OL) sequences of Vibrio parahaemolyticus were obtained from the National Center of Biotechnology Information Assembly database. The sequence similarity and polymorphism were analyzed by using blastn, fastANI, Sourmash and pixy tools. Recombination analysis was performed by using gubbins, and a molecular serotype prediction database of Vibrio parahaemolyticus was constructed.
    Results The O-antigen gene cluster (OL10V1) of Vibrio parahaemolyticus O10:K4 was significantly different from the typical O10 serotype gene cluster (OL10), but was most similar to OL4. In the 116 full-length sequences of OL10V1 in the public databases, only one single nucleotide mutation was found. The IS1634 and ISVa2 transposase genes carried by OL10V1 were widely distributed in Vibrio species, suggesting that this variant might formed through horizontal gene transfer (HGT).
    Conclusion This study revealed the potential molecular mechanism for the rapid spread of Vibrio parahaemolyticus O10:K4, and the constructed molecular serotype database can provide key technical support for genomic surveillance and traceability.
