2016-2023年湖北省荆门市H3N2亚型流感病毒血凝素基因特征分析

Genetic characterization of hemagglutinin gene of influenza A (H3N2) virus in Jingmen, Hubei, 2016−2023

    目的  了解2016—2023监测年度湖北省荆门市H3N2亚型流行性感冒(流感)病毒血凝素(HA)基因特征及抗原漂移情况。
    方法 对2016—2023监测年度湖北省荆门市分离到的60株H3N2流感毒株进行全基因组测序,利用MEGA 11、UCSF Chimera 1.18等软件对HA基因序列进行抗原变异分析和三维建模分析。
    结果  2016—2023监测年度共计5个监测年度有H3N2亚型流感病毒检出,共有3C.2a1、3C.2a2、3C.2a3、3C.2a1b.1b、3C.2a1b.2a.1a、3C.2a1b.2a.2a.3a.1共计6个进化簇流行,优势毒株从2016年的3C.2a2簇逐步变更为2023年的3C.2a1b.2a.2a.3a.1簇,两者在HA蛋白抗原表位上存在差异的氨基酸突变位点达21个,有I140K等13处氨基酸突变位点在表面模式结构图中差异明显。
    结论  2016—2023监测年度湖北省荆门市H3N2亚型流感病毒不断进化,发生了抗原漂移,应持续加强对H3N2亚型流感病毒的分子变异监测。

     

    Abstract:
    Objective To investigate the genetic characteristics of hemagglutinin (HA) and antigen drift of influenza A (H3N2) virus in Jingmen, Hubei Province.
    Methods A total of 60 strains of influenza A (H3N2) virus isolated in Jingmen from 2016 to 2023 were sequenced for whole genome. The antigenic variation and three-dimensional modeling of HA gene sequences were analyzed by using software MEGA 11 and UCSF Chimera 1.18.
    Results From 2016 to 2023, influenza A (H3N2) virus were detected in 5 years in surveillance, and a total of six evolutionary clusters, including 3C.2a1, 3C.2a2, 3C.2a3, 3C.2a1b.1b, 3C.2a1b.2a.1a, 3C.2a1b.2a.2a.3a.1 were detected. Among them, 3C.2a2, predominated in 2016, and 3C.2a1b.2a.2a.3a.1, predominated in 2023, had 21 amino acid mutation loci in HA protein epitope and 13 amino acid mutation loci, including I140K, showed significant differences in the surface pattern structure diagram.
    Conclusion Evolution and antigenic drift of influenza A (H3N2) virus occurred in Jingmen from 2016 to 2023. It is necessary to further strengthen the surveillance for molecular variation of influenza A (H3N2) virus.
