Analysis of whole genome evolution of influenza B virus Victoria lineage in Fujian, 2019−2022

    目的 研究2019—2022年福建省B型Victoria系流感病毒全基因组进化及分子特征。
    方法  选取福建省2019—2022年分离的48株B型Victoria系流感病毒毒株,全基因组扩增测序后用CLC Genomics Workbench 11.0软件拼接。 用MEGA 5.0软件对各节段进行系统进化树及分子特征分析。
    结果 与疫苗株B/Austria/1359417/2021相比,2019—2022年福建省毒株血凝素(HA)基因抗原决定簇和受体结合位点存在多个抗原决定簇(120环、150环、160环、190螺旋)和受体结合位点变异。 随着时间的推移HA糖基化位点数量减少,神经氨酸酶基因耐药位点及酶活性位点均保守。 聚合酶酸性蛋白基因、聚合酶碱性蛋白1基因、聚合酶碱性蛋白2基因、核蛋白基因、非结构蛋白基因、基质蛋白基因的系统进化树分支和进化距离平均值虽相对于HA变异情况更保守,但各有不同特点。
    结论 2019—2022年福建省B型流感病毒Victoria系流行株不同基因节段具有遗传多样性,可为疫苗或药物研发提供参考。

     

    Abstract:
    Objective To explore the complete genome evolution and molecular characteristics of influenza B virus Victoria lineage in Fujian province from 2019 to 2022.
    Methods A total of 48 strains of influenza B Victoria lineage isolated in Fujian province from 2019 to 2022 were selected, and software CLC Genomics Workbench 11.0 was used for segment joining after whole genome amplification sequencing. The phylogenetic and molecular characteristics of each segment were analyzed by using MEGA 5.0.
    Results The hemagglutinin (HA) gene of influenza B virus Victoria lineage isolated in Fujian during 2019−2022 had multiple antigen determinant (120 loop, 150 loop, 160 loop, 190 helix) and receptor binding site variants compared with vaccine strain B/Austria/1359417/2021. The number of HA glycosylation sites decreased over time. Neuraminidase gene resistance sites and enzyme active sites were conserved. The phylogenetic tree branches and evolutionary distances of polymerase acidic protein, polymerase basic protein 1, polymerase basic protein 2, nucleoprotein, nonstructural protein, matrix protein genes were more conservative than those of HA genes, but each had different characteristics.
    Conclusion There was genetic diversity in different gene segments of influenza B virus Victoria lineage circulating in Fujian from 2019 to 2022, which could provide reference for vaccine or drug development.
