3株抗菌药物活性评价用标准菌株的建立与评价

Establishment and evaluation of three standard strains for evaluating the activity of antimicrobial agents

    目的 参照《病原微生物菌(毒)种国家标准菌株评价技术标准》(WS/T 812-2022),在实验室条件下对保藏于中国医学科学院病原微生物菌(毒)种保藏中心药用微生物相关菌(毒)种保藏分中心的大肠埃希菌CCPM(A)-P-000101、肺炎克雷伯菌CCPM(A)-P-000102、金黄色葡萄球菌CCPM(A)-P-000100菌株,分别测定病原微生物学特征,推动这3株细菌成为抗菌药物活性评价中使用的标准株。
    方法 进行菌落形态和革兰染色观察,根据生化鉴定、抗生素敏感性测试、16S rDNA测序,以及多次传代后,观察和比较不同代次间细菌生长、药敏特性以及感染实验动物的100%最小致死量的差别,了解3株细菌的传代稳定性,明确其病原微生物学特征。
    结果 菌落形态、革兰染色、生化鉴定、16S rDNA测序结果证明CCPM(A)-P-000101、CCPM(A)-P-000102、CCPM(A)-P-000100菌株具备大肠埃希菌、肺炎克雷伯菌、金黄色葡萄球菌对应的典型特征,除对青霉素、氨苄西林等少数抗生素具有耐药性,3株细菌对多数受试抗生素具有敏感性表型。 3株细菌的G10菌株与G1菌株比较,生长曲线、药敏特征、感染实验小鼠的100%最小致死量均无显著差别,反映出它们的病原微生物学表型具有良好的传代稳定性。
    结论 大肠埃希菌CCPM(A)-P-000101、肺炎克雷伯菌CCPM(A)-P-000102、金黄色葡萄球菌CCPM(A)-P-000100菌株各自对应的菌种特征典型、稳定,适合作为标准株用于抗菌药物活性的评价等研究。

     

    Abstract:
    Objective To promote Escherichia coli CCPM(A)-P-0000101, Klebsiella pneumoniae CCPM(A)-P-0000102, and Staphylococcus aureus CCPM(A)-P-0000100, which were collected in the Division for Medicinal Microorganisms Related Strains, Chinese Academy of Medical Sciences (CAMS) Collection Center of Pathogenic Microorganisms, to become the standard strains used in the evaluation of antimicrobial activity, their microbiological characteristics were measured and identified according to the Technical Requirements for Evaluation of National Standard Strains of Pathogenic Microorganisms(WS/T 812—2022).
    Methods The bacterial morphology, biochemical testing, 16S rDNA sequencing, and susceptibility to antibiotics of the three strains were characterized by observation of the colonies on agar plates, Gram staining, and agar-dilution method. The passage stability of the three bacteria were further determined by comparing and analyzing the differences in bacterial growth curves, susceptibility to antibiotics, and 100% minimum lethal dose (MLD) to experimental mice between G10 and G1 strains.
    Results The colony morphology, Gram staining, biochemical testing, and 16S rDNA sequencing identified the strains CCPM(A)-P-0000101, CCPM(A)-P-0000102, and CCPM(A)-P-0000100 with typical characteristics corresponding to E. coli, K. pneumoniae, and S. aureus, respectively. Except for antibiotics such as penicillin and ampicillin, the three bacteria showed a susceptible phenotype to most of the tested antibiotics. There was no significant difference in growth curves, susceptibility to antibiotics, and 100% MLD to experimental animals between the G10 and G1 strain of the three bacteria, indicating that they have good passage stability.
    Conclusion The strains CCPM(A)-P-0000101, CCPM(A)-P-0000102, and CCPM(A)-P-0000100 own the typical and stable characteristics of their species, making them suitable as standard strains for the evaluation of antibacterial activity.
