高等级生物安全实验室终末消毒效果评价用枯草芽孢杆菌标准株的评价与应用

Evaluation and application of Bacillus subtilis standard strains for validation of terminal disinfection efficacy in high-level biosafety laboratories

  • 摘要:
    目的 依据《病原微生物菌(毒)种国家标准株评价技术标准》(WS/T 812—2022)建立高等级生物安全实验室终末消毒评价用枯草芽孢杆菌标准菌株。
    方法  选择4株国内分离的枯草芽孢杆菌,通过对标准菌株菌落形态、染色特性、生化特性以及全基因组测序,对标准菌株进行生物学特性和分子特征分析;再将4种标准菌株制成一定浓度的菌片,与市售的菌片一起对动物生物安全三级实验室及关键防护设备进行终末消毒效果的评价。
    结果 候选4株菌株均鉴定为枯草芽孢杆菌,全基因组均为环形,不含质粒;在相同消毒条件下,与ATCC9372一样,置于房间、生物安全柜和小鼠通风笼架4种候选菌株的菌片均可完全被杀灭。
    结论 本研究建立了高等级生物安全实验室终末消毒评价用枯草芽孢杆菌标准菌株,这些标准菌株的建立为消毒评价用菌株的国产化奠定了物质基础。

     

    Abstract:
    Objective To establish standard strains of Bacillus subtilis for evaluation of terminal disinfection in high-level biosafety laboratories according to the Technical Requirements for Evaluation of National Standard Strains of Pathogenic Microorganisms (WS/T 812—2022) .
    Methods Four strains of Bacillus subtilis isolated in China were used to study the colony morphology, staining characteristics, biochemical characteristics and whole genome sequencing of standard Strain, the biological and molecular characteristics of the standard strains were analyzed, and then the four standard strains were made into a certain concentration of bacterial tablets, evaluate the final disinfection efficacy of Animal Biosafety Level Ⅲ Laboratory and key protective equipment with commercially available bacterial tablets.
    Results All four candidate strains were identified as Bacillus subtilis, with a circular genome and no plasmids; Under the same disinfection conditions, like ATCC9372, the bacterial tablets of the four candidate strains placed in rooms, biosafety cabinets, and mouse ventilation cages can all be completely killed.
    Conclusion The standard strains of Bacillus subtilis for the evaluation of terminal disinfection in high-level biosafety laboratories were established in this study.
