一起札如病毒所致成年人急性胃肠炎暴发的分子流行病学研究

Molecular epidemiology of a cute gastroenteritis outbreak caused by Sapovirus in Adults

  • 摘要: 目的 了解一起成年人急性胃肠炎暴发流行的病原学。 方法 采用国家标准方法对样本进行沙门菌、志贺菌、金黄色葡萄球菌、溶血性链球菌、副溶血弧菌、变形杆菌、致泻性大肠埃希菌、蜡样芽孢杆菌、空肠弯曲菌及单核细胞增生李斯特菌10种致病菌进行检测。采用反转录-聚合酶链反应(RT-PCR)检测诺如病毒和札如病毒核酸,对阳性PCR产物进行核苷酸序列测定,并进行基因系统进化分析。 结果 7份粪便样本中4份为札如病毒核酸阳性。4株札如病毒与GⅠ. 2型参考株核苷酸序列同源性在94.7%~98.4%之间。基因进化分析显示,札如病毒深圳株与Sapovirus Hu/Oshima1/2009/JP、Houston/90(U95644)、Parkville/94/U73124 在同一进化树遗传分支,属于GⅠ. 2型。 结论 此起成年人急性胃肠炎暴发是由札如病毒GⅠ. 2型感染所致。

     

    Abstract: Objective To understand the etiology of a gastroenteritis outbreak in adults. Methods The stool samples were collected from the patients to conduct detections of Saimonella, Shigella, Staphylococcus aureus, Streptococcus hemolyticus, Vibrio parahaemolyticus, Bacillus proteus, Enteropathogenic Escherichia coli, Bacillus cereus, Campylobacter jejuni and Listeria monocytogenes according to national standard. Norovirus and Sapovirus nucleotide were detected by using RT-PCR, and PCR amplification products were cloned and sequenced. A phylogenetic tree was constructed by using Mega 3.1 software. Results Four of 7 diarrhea patients samples were Sapovirus positive, The homology between 4 strains of Sapovirus and reference strain of Sapovirus GⅠ.2 ranged from 94.7% to 98.4%. The phylogenetic analysis indicated that Shenzhen Sapovirus strain were in same cluster with Sapovirus Hu/Oshima1/2009/JP,Houston/90(U95644), Parkville/94/U73124 and belonged to GⅠ. 2. Conclusion The outbreak of gastroenteritis in adults was caused by Sapovirus GⅠ. 2.
