Analysis on PstⅠ digestion polymorphism of <I>omp2</I> gene of <I>Brucella melitensis</I> isolated in China

  • ObjectiveTo explore PstⅠ digestion polymorphism of Iomp2/I gene of IBrucella melitensis/I isolated in China. MethodsStandard strain of IBrucella melitensis/I, vaccine strain (M5), wild strain of IBrucella melitensis/I isolated in China and atypical strain were analyzed by polymerase chain reaction-restriction fragment length polymorphism analysis (PCR-RFLP). ResultsThe amplification product of Iomp2/I gene had two PstⅠ digestion pattern. Of the 46 strains analyzed, 43 had 2 straps of 238 bp and 44 bp, only 1 standard strain of IBrucella melitensis/I biotype 1 (16M) and 2 atypical strains of IBrucella melitensis/I biotype 1 isolated in Qinghai had 3 straps of 282 bp、238 bp and 44 bp. ConclusionPstⅠ digestion of Iomp2/I gene can distinguish some IBrucella melitensis/I in wild animal and IBrucella melitensis/I in cattle, which provide the evidence to reveal the geno-derivation of IBrucella./I
