First isolation and identification of human adenovirus type 14 in Liaoning

  • Objective To understand the genotype of 1 human adenovirus strain isolated from throat swab of a patient with acute respiratory disease. Methods The throat swabs were collected from ARD patients in Shenyang, Liaoning province and viruses were isolated by cell culture. The isolated virus causing cytopathogenic effect (CPE) on HEP-2 cells was tested by using polymerase chain reaction (PCR) with universal human adenovirus primers and the positive. PCR-product were sequenced and analyzed. The complete genome of Hexon were sequenced and compared with sequences of the other human adenovirus from GenBank. The phylogenetic tree of the complete genome of Hexon gene was constructed by using neighbor-joining method with software Mega 3.1. Results One adenovirus strain was isolated form a patient with ADR and the PCR result was positive. The sequence of amplifying segment was the same as human adenovirus type 14 reference strain. The entire genome of hexon was 2838 nucleotides long and was predicted to encode 946 amino acids, which shared 100% homology with human adenovirus type 14 reference strain. Phylogenetic analysis of the complete genome of Hexon showed the isolate and human adenovirus type 14 reference strain all belonged to one branch in the phylogenetic tree. Conclusion The strain isolated from the throat swab of a ARD patient in Shenyang was human adenovirus type 14 (GenBank accession number: KC825052).
