Correlation between KCNJ1 gene polymorphism and blood lipid level in Han ethnic group in Ningbo, Zhejiang

  • Objective To investigate the effect of single nucleotide polymorphisms (SNPs) rs675759, rs675388, rs2846679 of KCNJ1 gene on blood lipid level in Hanethnic group in Ningbo, Zhejiang province. Methods A total of 2 330 Han ethnic peopleaged 40 years in Yinzhou of Ningbo were recruited for this study. Epidemiological survey was conducted to collect their demographic and behavioral information, and blood samples were taken from them. PCR-LDR was performed to detect KCNJ1 rs675759, rs675388 and rs2846679 genotypes. Hardy-Weinberg was used to evaluate the genetic equilibrium. The effect of genotypes on the levels of total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) were evaluated by multivariable linear regression analysis, using gender, age, body mass index (BMI), and blood pressures as covariate. Results The successful genotyping rates for these SNPs were more than 99%, the differences were significant indicated by Hardy-Weinberg equilibrium test (P0.05). rs675759 decreased HDL-C by 0.12 mmol/L per G allele, rs675388 increased TC by 0.14 mmol/L and HDL-C by 0.12 mmol/L per T allele, rs2846679 decreased TC by 0.09 mmol/L and TG by 0.08 mmol/L per A allele. Conclusion KCNJ1 gene polymorphism was correlated with the levels of TC, TG and HDL-C in Han ethnic group in Ningbo.
